Alzheimer’s Disease is characterized by cognitive impairment, some causative factors being synaptic loss and defective neurogenesis. On a molecular level of progressive neurodegeneration, there is formation of amyloid-β(Aβ)-containing plaques and nerofibrillary tangles composed of hyperphoshorylated tau. The following diagram illustrates some proposed pathways which have been implicated in synaptotoxicity in Alzheimer’s Disease, driven either directly or indirectly by Aβ. The cause for most Alzheimer’s cases is still mostly unknown except for 1% to 5% of cases where genetic differences have been identifiled. Several competing hypotheses exist trying to explain the cause of the disease.
